YhbJ(RapZ) is an adaptor protein that recruits GlmZ to the catalytic domain of RNase E; GlmY is an anti-adaptor decoy sRNA that binds YhbJ(RapZ) competitively with GlmZ, protecting it from degradation (Göpel, 2013). GlmS synthesis is negatively feedback regulated by glucosamine-6-phosphate, and YhbJ(RapZ) enables degradation the GlmZ sRNA; unprocessed GlmZ upregulates glmS mRNA post-translationally by mediating an RNase E cleavage of the glmS mRNA (Kalamorz, 2007). The GlmY sRNA upregulates glmS post-transcriptionally (Urban, 2007). GlmS can be inactivated by various halomethyl and methyl ketone derivatives, of which bromomethyl ketone derivative is most effective because of its high binding affinity. Cys2 is an N-terminal nucleophile for GlmS.
