YdiV can act as an anti-FlhD4C2 factor in vitro, binding FlhD4C2 by binding FlhD and blocking FlhD4C2 activation of the flagellar regulon; YdiV of E. coli K-12 is poorly translated, explaining why E. coli K-12 does not have reduced motility at low nutrient concentrations mediated by YdiV and alleviated by fliZ mutation as observed for Salmonella (Wada, 2011; Wada, 2012). YdiV was named CdgR and characterized in Salmonella typhimurium wildtype strain ATCC 14028 as a protein required for the bacterium to resist host phagocyte oxidase in vivo; a Salmonella typhimurium ATCC 14028 cdgR(ydiV) mutant is reported to have seven-fold higher c-di-GMP levels (Hisert, 2005). However, in contrast to previously published results, a Salmonella typhimurium ATCC 14028 ydiV(cdgR,STM1344) mutant was found to have a lowered intracellular c-di-GMP and overexpression of ydiV(cdgR,STM1344) increased intracellular c-di-GMP, which may be attributable to differences in the c-di-GMP assay (soluble lysate vs total cell extraction); Salmonella typhimurium YdiV is a single domain EAL protein lacking phosphodiesterase, diguanylate cyclase, or c-di-GMP binding activity; YdiV(STM1344) is positive regulator of csgD transcription (Simm, 2009). In Salmonella typhimurium laboratory strain LT2 neither a ydiV mutation nor overexpression significantly affected c-di-GMP levels (Wada, 2011). Extracellular AI-1 stimulates ydiV RNA expression in an SdiA-dependent manner (Zhou, 2008). An sdiA ydiV double mutant has twofold reduced cAMP levels than inhibit quorum sensing system 2 (Zhou, 2008). ydiV RNA is 3.5-fold up-regulated by RpoS at ph 5 (Weber, 2006).
